- Burkholderia pseudomallei is a tropical pathogen and the causative agent of melioidosis, which displays a broad spectrum of symptoms and is therefore difficult to diagnose. Treating B. pseudomallei infections is challenging, as the bacterium is inherently resistant against multiple antibiotics. In this study, we investigate the effects of prostaglandin E2 (PGE2), a molecule known to inhibit antimicrobial activity in macrophages via EP2 and EP4 receptor signaling. Additionally, we examine the influence of the cannabinoid (CB) 2-arachidonoylglycerol (2-AG), which has been proposed to exhibit anti-inflammatory properties, on intracellular bacterial survival and pro-inflammatory, lytic cell death. We use bone-marrow-derived macrophages of wild type (WT) and ABHD6 knockout (KO) mice, which lack the serine hydrolase ABHD6 that processes 2-AG into a precursor of PGE2.
