Abstract:
Precursor lesions of pancreatic ductal adenocarcinoma (PDAC) and cholangiocellular carcinoma (CC) share many histological aspects and are graded similarly after the degree of dysplasia. Precursor lesions of PDAC are pancreatic intraepithelial neoplasia (PanIN) and papillary intraductal mucinous neoplasm (IPMN). Precursor lesions of CC are biliary intraepithelial neoplasia (BilIN) and intraductal papillary neoplasm of the bile duct (IPNB). Stepwise progression towards increasingly dysplastic lesions can be explained by molecular aberrations that shall be discussed in this thesis, with emphasis on gene mutations associated with malignant behavior. Literature on the most important genetic aberrations in the progression of PanIN, BilIN, IPMN and IPNB was collected using standard medical databases like PubMed. In the discussion part, emphasis was laid on differences in protein expression between biliary and pancreatic lesions. Furthermore, great importance was placed on differences in protein expression between high-grade lesions and low-grade lesions and between non-invasive high-grade lesions and invasive carcinomas of the respective organ-system.
