Abstract:
HLA-G is a major human histocompatibility complex class Ib gene product with limited polymorphism and is expressed in the extravillous cytotrophoblast of the human placenta. This molecule plays a central role in maternal tolerance of the semiallogenic fetus. To date seven different HLA-G isoforms have been described, namely HLA-G1, -G2, -G3 and G4 membrane-bound and HLA-G5, -G6 and G7 soluble proteins, which are generated by translation of alternatively spliced mRNA. In this study monoclonal antibodies were produced by immunizing Balb/c mice with synthetic peptides corresponding to selected amino acid sequences of the HLA-G molecule. The splenocytes of his mice were fused with mouse myeloma cells. The culture supernatants of the hybridomas were tested by ELISA for antibody production and then further investigations were performed by Immunohistochemistry and Western Blotting.
