The impact of a non-selective endothelin-antagonist (Bosentan) on the hepatic microcirculation after warm ischemia

- There is evidence that endothelin-1 (ET-1) is involved in disturbances of the hepatic microcirculation after warm ischemia. In this study we investigated the influence of a non-selective endothelinreceptor-antagonist (Bosentan) on ischemia/reperfusion damage of the liver by means of intravital fluorescence microscopy (IVM). Clamping of the left liver lobe was performed in 12 male Wistar rats for 70 min. The treatment group (n = 6) received 15 mg Bosentan per kg body weight 1 min prior to reperfusion. Controls (n = 6) received an equivalent amount of Ringer's solution. Between 20 - 90 min after reperfusion the heptic microcirculation was studied. Application of Bosentan reduced the number of non-perfused sinusoids and adherent leukocytes in sinusoids and venules. Sinusoidal diameters increased after treatment with Bosentan. Our results imply a beneficial impact of Bosentan on hepatic microhemodynamics after warm ischemia in rat livers.
